Angiotensin II (ANGⅡ)
Angiotensin II is a major vasoactive peptide that is locally produced and released into the bloodstream. Angiotensin Ⅱ hormone, can adjust water and salt metabolism and blood pressure on cardiovascular, kidney disease, endocrine disease diagnosis and treatment is important. Angiotensin Ⅱ renin - angiotensin system is the main active substances. It not only plays a key role in the acute and chronic regulation of systemic arterial blood pressure, but also is an important regulator of cardiovascular function. Angiotensin Ⅱ is specific to participate in the course of the disease and thus become the ideal target of drug therapy. The change of the determination of angiotensin Ⅱ, is essential to study drug curative effect.
Recently, researchers at the cochan institute in Paris set out to complete a meaningful study in which they found that angiotensin II promotes cancer cell migration. In the study, they used an in vivo model of cancer metastases. To study important steps in cancer metastasis, they injected a stable fluorescent breast cancer cell line (D3H2LN) into nude mice. Through real-time live imaging studies, they found that angiotensin II promoted the formation of metastatic foci.
It was found that after pretreatment with the polypeptide, the number of mice with metastatic cancer cells increased, and the number and size of metastatic tumors in each mouse increased accordingly. In vitro, angiotensin II promotes cancer metastasis by facilitating the adhesion of cancer cells to endothelial cells, transendothelial migration, and through the extracellular matrix. At the molecular level, DNA microarray analysis showed that there were 102 genes of differential expression after pretreatment of angiotensin II. Angiotensin II regulates two genes closely related to its precursor angiotensinogen. In these genes, up-regulated expression of MMP2/MMP9 and ICAM1M was associated with a network of genes involved in cell adhesion, migration, and invasion. All of these suggest that targeting angiotensin II may be an effective strategy for inhibiting invasive breast cancer metastasis.
